Oxidized Forms

Oxidized forms are molecules or ions that have undergone oxidation, typically by losing electrons, losing hydrogen, or gaining oxygen, and they are central to biological redox chemistry. They arise when an oxidizing agent accepts electrons from a reduced partner, creating a redox pair whose interconversion can transfer energy or alter molecular activity. In biology, oxidized forms such as NAD+ and oxidized glutathione support metabolism and cellular defense, while oxidation of proteins, lipids, and nucleic acids can influence signaling or contribute to oxidative damage. Studying these forms helps researchers interpret electron transport, redox regulation, disease mechanisms, and biochemical assay results.

Rapid Quantification of Oxidized and Reduced Forms of Glutathione Using Ortho -phthalaldehyde in Cultured Mammalian Cells In Vitro

Quantification of both oxidized and reduced forms of glutathione (GSSG and GSH, respectively) has been achieved through the use of Ortho-phthalaldehyde (OPA). OPA becomes highly fluorescent once conjugated to GSH but is unable to conjugate GSSG until reduced. Here, we describe a multiparametric assay to quantify both using protein quantification for normalization.
