Oxygen Loading Unloading

Oxygen loading and unloading is the reversible uptake of oxygen by hemoglobin in red blood cells and its release to body tissues, a process essential for aerobic metabolism. In pulmonary capillaries, high oxygen partial pressure promotes hemoglobin binding; in metabolically active tissues, lower oxygen pressure and increased carbon dioxide, acidity, and temperature reduce hemoglobin’s affinity, encouraging oxygen release through the Bohr effect. Understanding this exchange explains how blood supports cellular respiration and adapts to exercise, altitude, and disease, while informing research on respiratory physiology, anemia, blood substitutes, and disorders that impair oxygen delivery.

Microelectrode-Mediated FM Dye Loading and Unloading in Drosophila Larvae Nerve Cells

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.

We developed an alternant hindlimb unloading model in mice. The primary advantage of our hindlimb unloading tail-ring method over the conventional Morey-Holton tail-traction technique is a simple straightforward procedure that minimizes stress upon the animal.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
