Oxygen Mapping

Oxygen mapping is the measurement and visualization of oxygen concentration across a biological sample, tissue, or cellular environment. It uses oxygen-sensitive probes, dyes, or sensors whose fluorescence, phosphorescence, or signal intensity changes when oxygen interacts with the indicator, allowing spatial gradients and local oxygen levels to be estimated. In biology, these maps reveal how oxygen availability varies within tumors, developing tissues, organoids, and engineered cultures, where diffusion, cellular respiration, and blood flow create heterogeneous microenvironments. Oxygen mapping supports studies of metabolism, hypoxia, tissue function, and disease progression, while helping researchers evaluate how cells respond to changing oxygen conditions.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
