Pancreatic Xenografts

Pancreatic xenografts are in vivo models created by implanting pancreatic tumor cells or tissue from one species, commonly human, into an immunocompromised animal, allowing cancer biology to be studied in a living system. Because the host immune response is suppressed, the graft can establish, vascularize, and grow, while its placement as a subcutaneous or orthotopic implant influences tumor behavior and surrounding tissue interactions. These models help researchers examine pancreatic tumor growth, invasion, metastasis, and responses to anticancer treatments. By preserving features of human disease more closely than many simplified cell assays, pancreatic xenografts support preclinical drug evaluation and investigation of tumor biology.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.

This video describes the technique of DCE-MRI for orthotropic pancreatic tumor xenografts in mice. DCE-MRI is a non-invasive method to analyze microvasculature in a target tissue, and useful to assess vascular response in a tumor following a novel therapy. This method will assist investigators to apply DCE-MRI for orthotopic gastrointestinal cancer mouse models.

This protocol describes optimization procedures in a virus-based dual fluorescence-labeled tumor xenograft model using larval zebrafish as hosts. This heterogeneous xenograft model mimics the tissue composition of pancreatic cancer microenvironment in vivo and serves as a more precise tool for assessing drug responses in personalized zPDX (zebrafish patient-derived xenograft) models.

We present a protocol to generate a minimally invasive orthotopic pancreatic cancer model by ultrasound-guided injection of human pancreatic cancer cells and the subsequent monitoring of tumor growth in vivo by ultrasound imaging.
