Perivascular Elastase Application

Perivascular elastase application is an experimental method that applies the proteolytic enzyme elastase around a blood vessel to induce localized vascular remodeling, commonly as a model of abdominal aortic aneurysm. Elastase degrades elastin in the vessel wall, weakening its structural support and promoting inflammation, extracellular matrix remodeling, and progressive vessel dilation. In biology research, this approach helps investigators examine aneurysm development, interactions among vascular cells and matrix components, and mechanisms that contribute to vascular wall instability. It also provides a controlled platform for evaluating genetic factors, disease pathways, imaging methods, and potential therapies for vascular disorders.

Human perivascular stem cells (PSCs) are a novel stem cell class for skeletal tissue regeneration similar to mesenchymal stem cells (MSCs). PSCs can be isolated by FACS (fluorescence activated cell sorting) from adipose tissue procured during standard liposuction procedures, then combined with an osteoinductive scaffold to achieve bone formation in vivo.

The model has the ability to add broad insight into the pathobiology of AAA due to the emergence of numerous transgenic and gene knockout mice.
