Phalloidin Labeling

Phalloidin labeling is a fluorescence-based method for visualizing filamentous actin (F-actin), a major cytoskeletal component that supports cell shape, movement, and organization. Phalloidin, a cyclic peptide derived from poisonous Amanita mushrooms, binds selectively and tightly to F-actin; when linked to a fluorophore, it marks actin filaments for detection by fluorescence or confocal microscopy. In biology, this technique reveals cytoskeletal architecture, cell morphology, and changes in actin organization during processes such as migration, adhesion, division, and differentiation. It is widely used to compare cellular structures and assess how experimental treatments affect the actin cytoskeleton.
