Phosphatase Enzymes

Phosphatase enzymes are biological catalysts that remove phosphate groups from proteins, sugars, nucleotides, and other molecules, helping regulate cellular chemistry. They promote dephosphorylation through hydrolysis, using water to break phosphate ester or phosphoanhydride bonds; many phosphatases also rely on metal ions or conserved catalytic residues to stabilize reaction intermediates. In biology, these enzymes balance the activity of kinases and control processes such as signal transduction, metabolism, cell-cycle progression, and gene regulation. Their specificity and regulatory roles make phosphatases important subjects in biochemical research and potential targets for understanding or treating cellular dysfunction.

Multi-enzyme Screening Using a High-throughput Genetic Enzyme Screening System

This work presents a method of high-throughput screening using a universal genetic enzyme screening system that can be theoretically applied to over 200 enzymes. Here, the single screening system identifies three different enzymes (lipase, cellulase, and alkaline phosphatase) by simply changing the substrate used (p-nitrophenyl acetate, p-nitrophenyl-β-D-cellobioside, and phenyl phosphate).
