Photoreceptor Photo Response

Photoreceptor photoresponse is the change in electrical and chemical signaling that occurs when a rod or cone in the retina detects light, providing the first step in vision. In darkness, cGMP keeps cation channels open; photon absorption activates an opsin and a G protein cascade that stimulates phosphodiesterase, lowers cGMP, closes these channels, and hyperpolarizes the cell, reducing glutamate release. Comparing photoresponses across rods and cones helps explain sensitivity, color vision, adaptation, and signal transmission to retinal circuits, while measurements of these responses support research on retinal physiology, inherited visual disorders, and therapies designed to restore light detection.

Sharp microelectrodes enable accurate electrophysiological characterization of photoreceptor and visual interneuron output in living Drosophila. Here we show how to use this method to record high-quality voltage responses of individual cells to controlled light stimulation. This method is ideal for studying neural information processing in insect compound eyes.
