Plant Cell Isolation

Plant cell isolation is a laboratory technique used to separate individual cells from plant tissues, enabling their structure, physiology, and responses to be studied under controlled conditions. Researchers commonly remove the cell wall through enzymatic digestion with cellulase and pectinase, while an osmotic stabilizer helps prevent the resulting protoplasts from bursting. Isolated cells and protoplasts support investigations of membrane function, cell signaling, gene expression, and plant-pathogen interactions, and they can also be used for genetic transformation, hybridization, and tissue culture. By reducing complex tissues to accessible cellular systems, plant cell isolation connects whole-plant biology with cellular and molecular research.

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.

Measuring the osmotic water permeability coefficient (Pf) of cells can help understand the regulatory mechanisms of aquaporins (AQPs). Pf determination in spherical plant cell protoplasts presented here involves protoplasts isolation and numerical analysis of their initial rate of volume change as a result of an osmotic challenge during constant bath perfusion.

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.
