Podocyte Marker Expression

Podocyte marker expression refers to the production and detection of molecular proteins or transcripts that identify podocytes, specialized cells that support the kidney’s glomerular filtration barrier. Markers such as nephrin, podocin, synaptopodin, and WT1 reflect podocyte identity and specialized structure, and researchers measure them using immunofluorescence, immunohistochemistry, western blotting, or RT-qPCR. Changes in marker abundance or localization can indicate podocyte differentiation, injury, dedifferentiation, or loss, often alongside disruption of the slit diaphragm and cytoskeleton. In biology and renal research, these measurements help characterize glomerular development, investigate proteinuric diseases, evaluate experimental models, and assess potential therapies aimed at preserving filtration function.

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

The present protocol allows the recovery of tube-like structures formed in the classical in vitro angiogenesis-tube formation assay where cells, such as trophoblast cells, organize in capillary-like structures over a layer of extracellular basement membrane matrix. The recovered structures are used for RNA and protein isolation for various biochemical analyses.
