Polystyrene Coating Oxygen Plasma

Polystyrene coating oxygen plasma is a surface-treatment method that modifies polystyrene to improve its suitability for biological experiments. During treatment, an ionized oxygen gas produces reactive species that interact with the polymer surface, breaking chemical bonds and introducing oxygen-containing functional groups; this increases surface wettability and can alter protein adsorption. In biology, treated polystyrene supports more consistent coating of extracellular matrix proteins and attachment of cultured cells, making it useful for cell culture, microscopy, and assays that depend on controlled cell-surface interactions. Such treatment can improve reproducibility while preserving the underlying polymer for laboratory studies of adhesion, growth, and cellular responses.

An experimental setup was created for the helium-operated kHz frequency plasma jet. The setup includes a cage for the plasma power supply and jet and an in-house built reactor to monitor plasma-induced reactive species without the interference of the ambient atmosphere.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
