Pupal Retina Dissection

Pupal Retina Dissection is a laboratory technique for isolating developing retinal tissue from an insect pupa so its cellular organization and maturation can be examined. Under a stereomicroscope, the cuticle and surrounding tissues are carefully removed, and the delicate retina is separated with fine instruments before fixation, antibody staining, or microscopy; controlled handling helps preserve retinal architecture. This preparation supports analysis of photoreceptor differentiation, cell patterning, neuronal connectivity, and gene or protein localization during development. In biology research, it provides a direct way to relate developmental mechanisms to retinal structure and evaluate genetic or experimental perturbations.

The Drosophila retina is a crystal-like lattice composed of a small number of cell types that are generated in a stereotyped manner 1. Its amenability to sophisticated genetic analysis allows the study of complex developmental programs. This protocol describes dissections and immunohistochemistry of retinas at three discrete developmental stages, with a focus on photoreceptor differentiation.

The ability to accurately detect transcripts or proteins in Drosophila tissues is critical for studying their abundance and localization related to the development process. Here is the description of a straightforward procedure to dissect pupal wings. These wings can be used as samples in several techniques (immunohistochemistry, PCR assay, etc.).

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
