Recombinant Protein Expression

Recombinant protein expression is the production of a selected protein by introducing its encoding DNA into a host cell, allowing researchers to obtain defined proteins for study or use. In a typical workflow, a gene is inserted into an expression vector and transferred into bacteria, yeast, insect, or mammalian cells; host transcription and translation then generate the protein, which may be collected and purified from cells or culture medium. Researchers can adjust promoters, culture conditions, and host systems to improve yield, folding, or post-translational modification. The method supports protein structure and function studies, antibody and enzyme production, drug development, diagnostics, and biotechnology manufacturing.

Expression of Recombinant Proteins in the Methylotrophic Yeast Pichia pastoris

The protocol describes protein expression using the methylotrophic yeast Pichia pastoris. The preparation of electrocompetent yeast cells, transformation of the vector with the gene of interest into P. pastoris and yeast DNA purification are also performed. Western blot analysis and protein purification build the last steps in this protein expression protocol.

Here we describe a way to express correctly folded and functional influenza virus surface antigens derived from the novel Chinese H7N9 virus in insect cells. The technique can be adapted to express ectodomains of any viral or cellular surface proteins.

Plants offer a novel system for the production of pharmaceutical proteins on a commercial scale that is more scalable, cost-efficient and safe than current expression paradigms. In this study, we report a simple and convenient, yet scalable approach to introduce target-gene containing Agrobacterium tumefaciens into plants for protein transient expression.

Nonlytic insect cell expression systems are underutilized for production, cellular trafficking/localization, and recombinant protein functional analysis. Here, we describe methods to generate expression vectors and subsequent transient protein expression in commercially available lepidopteran cell lines. The co-localization of Bemisia tabaci aquaporins with subcellular fluorescent marker proteins is also presented.
