Replica Plating

Replica plating is a microbiological technique that copies the spatial arrangement of colonies from a master agar plate onto one or more fresh plates, allowing researchers to compare the same cell populations under different conditions. A sterile velvet or similar surface picks up cells from each colony and transfers them to new media while preserving their relative positions. By comparing growth patterns, scientists can identify auxotrophic mutants, antibiotic-resistant cells, or organisms affected by specific nutrients or environmental conditions. This method links colony phenotype to genotype and remains useful in bacterial genetics, microbial physiology, selection experiments, and screening for mutations.

Fabrication, Densification, and Replica Molding of 3D Carbon Nanotube Microstructures

We present methods for fabrication of patterned microstructures of vertically aligned carbon nanotubes (CNTs), and their use as master molds for production of polymer microstructures with organized nanoscale surface texture. The CNT forests are densified by condensation of solvent onto the substrate, which significantly increases their packing density and enables self-directed formation of 3D shapes.

This article illustrates how the expression of neurotransmitter receptors can be quantified and the pattern analyzed at synapses with identified pre and postsynaptic elements using a combination of viral transduction of optogenetic tools and the freeze-fracture replica immunolabeling technique.

Aseptic Laboratory Techniques: Plating Methods

When working with media and reagents used to culture microorganisms, aseptic technique must be practiced to ensure contamination is minimized. A variety of plating methods are routinely used to isolate, propagate, or enumerate bacteria and phage, all of which incorporate procedures that maintain the sterility of experimental materials.

Here we describe the Replica Set method, an approach to quantitatively measure C. elegans lifespan/survival and healthspan in a high-throughput and robust manner, thus allowing screening of many conditions without sacrificing data quality. This protocol details the strategy and provides a software tool for analysis of Replica Set data.
