Reporter Signal

A reporter signal is a measurable biological output that indicates the activity, location, or timing of a gene, promoter, pathway, or cellular process. It is generated when a reporter gene, such as one encoding a fluorescent, luminescent, or color-producing protein, is expressed under the control of a selected regulatory sequence; the resulting signal can then be detected and quantified. In biology, reporter signals help researchers monitor gene expression, trace cell behaviors, assess signaling pathways, and evaluate experimental treatments. These readouts support studies of regulation and function while enabling comparisons across cells, tissues, and experimental conditions.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

We describe the procedure and data analysis of a chemical screening system for glucocorticoid stress hormone signaling using zebrafish larvae: the Glucocorticoid Responsive In vivo Zebrafish Luciferase activitY (GRIZLY) assay. The assay sensitively and specifically detects effects on glucocorticoid signaling by compounds that require metabolization or affect endogenous glucocorticoid production.
