Reporter Transgenes

Reporter transgenes are engineered DNA constructs that link a detectable reporter gene to regulatory sequences, allowing researchers to visualize or measure gene activity in living cells, tissues, or organisms. When the regulatory elements respond to specific transcription factors or cellular conditions, they drive expression of reporters such as fluorescent proteins or enzymes, producing a signal that reflects the timing, location, or level of pathway activation. In biology, reporter transgenes help map developmental patterns, analyze promoter function, monitor signaling pathways, and evaluate genetic or environmental effects. Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

Transgenic worms are commonly used in C. elegans research. Described is a simple, yet effective, protocol to introduce transgenes into worms using biolistic bombardment with DNA-coated gold particles. The effort involved and results of bombardment compare favorably with microinjection for the generation of transgenic animals.

Differentiating Non-Transgenic and Transgenic Human Pluripotent Stem Cells into Neural Progenitor Cells

This video demonstrates the generation of neural progenitor cells from non-transgenic and transgenic human pluripotent stem cells (hPSCs). hPSCs are differentiated into neural progenitor cells using a neuronal differentiation medium. While differentiation is induced in non-transgenic cells using small molecules, the transgenic hPSCs are differentiated by adding an antibiotic that induces the promoter for a neuronal differentiation-specific transcription factor.
