Respiratory Syncytial Virus

Respiratory syncytial virus (RSV) is an enveloped, negative-sense single-stranded RNA virus that infects the respiratory tract and is a major cause of respiratory illness, especially in infants and older adults. Its fusion (F) protein helps the virus enter respiratory epithelial cells and promotes the formation of multinucleated cell clusters called syncytia, while viral replication and host inflammation can obstruct small airways. Studying RSV biology clarifies viral entry, replication, immune responses, and disease progression. This knowledge supports the development of vaccines, monoclonal antibodies, antiviral therapies, and improved strategies for preventing severe respiratory disease.

We describe a method for generating and amplifying genetically modified respiratory syncytial viruses (RSVs) and an optimized plaque assay for RSVs. We illustrate this protocol by creating two recombinant viruses that respectively allow quantification of RSV replication and live analysis of RSV inclusion bodies and inclusion bodies-associated granules dynamics.
