Retrieval Signal

A retrieval signal is a molecular sequence or structural feature that directs a cellular component back to its proper compartment after it has moved elsewhere, helping maintain organelle identity and protein distribution. In the secretory pathway, for example, the KDEL sequence on soluble endoplasmic reticulum proteins is recognized by KDEL receptors in the Golgi, which package these proteins into COPI-coated vesicles for return to the endoplasmic reticulum. Studying retrieval signals clarifies how cells preserve compartment-specific functions, regulate protein trafficking, and prevent the loss or mislocalization of essential proteins. These mechanisms are also important for investigating cellular stress, secretion, and diseases linked to trafficking defects.
