Reverse Transcriptase

Reverse transcriptase is an enzyme that synthesizes DNA from an RNA template, reversing the usual flow of genetic information and providing a foundation for studying RNA biology. The enzyme binds RNA, uses a primer and deoxynucleotides to build a complementary DNA (cDNA) strand through base pairing, and some reverse transcriptases also degrade the RNA strand through RNase H activity. In molecular biology, this process supports reverse-transcription PCR, cDNA library construction, gene-expression analysis, and detection of RNA viruses. Reverse transcriptase also plays a central role in retroviral replication and helps researchers examine how RNA is converted into stable, analyzable DNA.

Increasing cDNA Yields from Single-cell Quantities of mRNA in Standard Laboratory Reverse Transcriptase Reactions using Acoustic Microstreaming

We describe a novel method for increasing cDNA yield from single-cell quantities of mRNA in otherwise standard laboratory reverse transcription reactions. The novelty resides in the use of a micromixer, which utilizes the phenomenon of acoustic microstreaming, to mix fluids at microliter scales more effectively than shaking, vortexing or trituration.

Human telomerase reverse transcriptase (TERT) synthesizes not only telomeric DNA but also double-stranded RNA through RNA-dependent RNA polymerase activity. Here, we describe a newly established assay to detect RNA-dependent RNA polymerase activity of endogenous TERT.
