Rfp Pneumococci

Rfp pneumococci are Streptococcus pneumoniae cells engineered to produce red fluorescent protein (RFP), making the bacteria visible through fluorescence-based imaging and analysis. The rfp gene directs synthesis of a fluorescent protein whose internally formed chromophore emits red light after excitation, allowing labeled pneumococci to be tracked without relying solely on stains or antibody-based detection. This approach supports studies of pneumococcal growth, localization, host-cell interactions, and biofilm organization in biology and microbiology. By linking bacterial behavior to observable fluorescence, RFP labeling helps researchers monitor infection-related processes and evaluate experimental conditions with greater spatial and temporal resolution.

We describe a novel red fluorescent protein (RFP) reporter that is expressed specifically in the Drosophila eye. We detail a methodology for dissection of the eye imaginal disc and how this reporter can be used to aid in the dissection and identification of specific cell types in the developing eye.

Source: Jagau, H. et al. Pneumococcus Infection of Primary Human Endothelial Cells in Constant Flow. J. Vis. Exp. (2019)This video demonstrates the real-time visualization of pneumococcal attachment to Von Willebrand Factor (VWF) strings on endothelial cells under constant media flow conditions. It outlines the steps for stimulating VWF release, labeling the strings, and quantifying bacterial binding.
