Ribonucleoprotein Complex

A ribonucleoprotein complex is a molecular assembly composed of RNA and one or more proteins, enabling cells to organize, protect, and regulate genetic information. Its function depends on specific RNA–protein interactions, in which proteins recognize RNA sequences or structures and, together with the RNA, form functional units for RNA processing, transport, translation, or degradation. Examples include ribosomes, spliceosomes, and telomerase, each supporting essential steps in gene expression and genome maintenance. Studying ribonucleoprotein complexes helps explain how cells control RNA fate and provides insight into diseases caused by disrupted RNA–protein interactions, as well as opportunities for molecular diagnostics and therapeutic design.

Influenza viruses replicate their RNA genome in association with host-cell chromatin. Here, we present a method to purify intact viral ribonucleoprotein complexes from the chromatin of infected cells. Purified viral complexes can be analyzed by both Western blot and primer extension of protein and RNA content, respectively.
