Scaffold Materials

Scaffold materials are biomaterials used to create three-dimensional frameworks that support cells, tissues, or engineered biological constructs, making them central to tissue engineering and regenerative biology. Their architecture, porosity, surface chemistry, mechanical properties, and degradation behavior determine how cells attach, migrate, receive nutrients, and organize as the scaffold gradually integrates with or is replaced by new tissue. Materials may be natural, synthetic, or hybrid, and can be shaped into hydrogels, porous matrices, or fibrous structures. By matching biological and mechanical requirements, scaffold materials enable controlled cell culture, tissue repair strategies, and models for studying development, disease, and therapeutic responses.

2016

In this work, poly(lactic acid)/polyethylene glycol (PLA/PEG) scaffolds were prepared by using a combination of melt mixing and selective leaching. The method herein discussed permitted to develop three-layer scaffolds by highly controlling both porosity and pore size. The mechanical properties were also evaluated in a physiological environment.

Scaffolds capable of fitting within cranio-maxillofacial (CMF) bone defects while exhibiting osteoconductivity and bioactivity are of interest. This protocol describes the preparation of a shape memory scaffold based on polycaprolactone diacrylate (PCL-DA) using a solvent-casting particulate-leaching (SCPL) method employing a fused salt template and application of a bioactive polydopamine coating.

This protocol combines electrospinning and microspheres to develop tissue engineered scaffolds to direct neurons. Nerve growth factor was encapsulated within PLGA microspheres and electrospun into Hyaluronic Acid (HA) fibrous scaffolds. The protein bioactivity was tested by seeding the scaffolds with primary chick Dorsal Root Ganglia and culturing for 4-6 days.
