Seed Culture Expansion

Seed culture expansion is the stepwise growth of a small, actively growing culture into larger volumes, providing sufficient biological material for experiments, production, or downstream analysis. The process begins with a healthy starter culture that is transferred into fresh, nutrient-containing medium under conditions suited to the organism or cell type, allowing population growth while maintaining viability and desired characteristics. In biology, seed culture expansion supports reproducible cell and microbial cultivation, improves the consistency of inocula, and supplies material for assays, bioprocessing, and larger-scale culture systems. Careful control of transfer timing, contamination, and culture conditions helps preserve culture quality and experimental reliability.

Source: J. Jacob Chavez, Ryan T. Davis, and Taylor D. Sparks, Department of Materials Science and Engineering, The University of Utah, Salt Lake City, UT Thermal expansion is extremely important when considering which materials will be used in systems that experience fluctuations in temperature. A high or low thermal expansion in a material may or may not be desirable, depending on the application. For instance, in a common liquid thermometer, a material with a high thermal expansion would be...

Seed Structure and Early Development of the Sporophyte

2020

Seed structures are composed of a protective seed coat surrounding a plant embryo, and a food store for the developing embryo. The embryo contains the precursor tissues for leaves, stem, and roots. The endosperm and cotyledons—seed leaves—act as the food reserves for the growing embryo. The embryo contains a double set of chromosomes, one set from each parent. Fertilization of the haploid egg by the haploid sperm gives rise to the zygote, which develops into the embryo. The endosperm is a...

Isolation of Tomato Seed-Associated Microbiota Through Controlled Germination and Comparative Culturing

2026

This protocol combines standardized seed surface sterilization with controlled germination to enable the reproducible isolation of tomato seed-associated microbiota during the seed-to-seedling transition. The approach enhances recovery of culturable microorganisms that are underrepresented in dry seeds and supports comparative culturomic studies during early seedling development.

A Seed Coat Bedding Assay to Genetically Explore In Vitro How the Endosperm Controls Seed Germination in Arabidopsis thaliana

We present the procedure to assemble a seed coat bedding assay (SCBA) from Arabidopsis thaliana seeds. The SCBA was shown to be a powerful tool to explore genetically and in vitro how the endosperm controls seed germination in dormant seeds and in response to light cues. The SCBA is in principle applicable under any situation where the endosperm is suspected to influence embryonic growth.
