Single Cell Digestion

Single-cell digestion is the process of converting a multicellular tissue or cell aggregate into a suspension of individual, viable cells for biological analysis. It typically combines enzymatic cleavage of extracellular matrix and cell-adhesion proteins, using enzymes such as collagenase or trypsin, with controlled mechanical trituration; incubation time, temperature, and agitation must be optimized to limit cell damage and preserve surface markers. The resulting suspension supports cell counting, flow cytometry, single-cell sequencing, primary culture, and studies of tissue heterogeneity, enabling researchers to link molecular profiles to distinct cell types. Careful digestion improves yield and viability, while incomplete dissociation or excessive treatment can bias downstream measurements and obscure rare populations.
