Single Cell Population

A single cell population is a group of individual cells examined separately to reveal differences that bulk measurements can obscure, making it valuable for understanding biological heterogeneity. Researchers isolate or partition cells, then measure features such as gene expression, surface proteins, morphology, or cellular function using single-cell assays; computational clustering can organize cells with shared characteristics into subpopulations. In biology, single cell population analysis helps identify rare cell types, define developmental or disease-related states, and trace changes in tissues over time. These data support cell atlas construction, biomarker discovery, and more precise models of normal and pathological processes.

Studying Proteolysis of Cyclin B at the Single Cell Level in Whole Cell Populations

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.

Antibiotic persistence describes the ability of small subpopulations within a sensitive isogenic population to transiently tolerate high doses of bactericidal antibiotics. The present protocol combines approaches to characterize the antibiotic persistence phenotype at the molecular and cellular levels after exposing Escherichia coli to lethal doses of ofloxacin.
