Single-worm Quantification

Single-worm quantification is the measurement of morphological, behavioral, developmental, or physiological traits in individual worms, providing resolution that population-level averages can obscure. The approach typically isolates or identifies each animal, records defined features through microscopy, imaging, or direct observation, and converts those measurements into standardized numerical data for comparison across individuals and conditions. In biology, single-worm quantification supports analysis of phenotypic variation, genotype or treatment effects, growth, movement, and survival. It can reveal rare or heterogeneous responses, strengthen reproducibility, and help connect individual-level traits with genetic, cellular, and environmental mechanisms.

Screening for mutants with phenotypic defects is a straightforward method for identifying genes that function in a given biological process. In this article we describe how to culture free living worms (e.g., Pristionchus pacificus) in the laboratory and show two different mutagenesis methods, EMS and TMP/UV.

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Introduction to C. elegans The nematode (roundworm) Caenorhabditis elegans, commonly known as C. elegans, is a widely used non-vertebrate animal model for studying fundamental biological processes. Because many cellular and...

Source: Morgan, E., et al. Selective Cleaning of Wild Caenorhabditis Nematodes to Enrich for Intestinal Microbiome Bacteria. J. Vis. Exp. (2021)This video demonstrates a protocol for isolating intestinal bacteria of interest from wild Caenorhabditis worms. The worms are first starved and washed to eliminate residual feeding bacteria from their surface and intestinal lumen while preserving the target microbes. After surface cleaning and multiple crawling steps, the worms are dissected to extract...

Mechanical Disruption of Individual Worms to Assess Variability in Host-Bacteria Interactions

Source: Taylor, M. N. et al. Using Single-Worm Data to Quantify Heterogeneity in Caenorhabditis elegans-Bacterial Interactions. J. Vis. Exp. (2022)This video demonstrates the quantification of intestinal bacterial load variability in individual Caenorhabditis elegans through mechanical disruption. It outlines the steps for sample preparation, mechanical disruption, serial dilution, and colony counting to assess differences in intestinal bacterial load among individual worms.
