Sodium Currents

Sodium currents are ionic flows carried by sodium ions across cell membranes, and they are essential for electrical signaling in neurons, muscle cells, and other excitable tissues. When membrane depolarization reaches a threshold, voltage-gated sodium channels open rapidly, allowing Na+ to enter down its electrochemical gradient; channel inactivation then stops the inward current and supports repolarization. Measuring sodium currents with patch-clamp electrophysiology helps researchers characterize action potentials, channel kinetics, and the effects of mutations or drugs. These studies clarify how nervous and cardiovascular systems function and support research into epilepsy, pain, cardiac arrhythmias, and channel-targeting therapies.

Demonstration of Proteolytic Activation of the Epithelial Sodium Channel (ENaC) by Combining Current Measurements with Detection of Cleavage Fragments

Proteolytic activation of the epithelial sodium channel (ENaC) heterologously expressed in Xenopus laevis oocytes can be demonstrated by combining current measurements with a biotinylation approach to investigate the appearance of ion channel cleavage products at the cell surface. Functionally important cleavage sites can be identified by using site-directed mutagenesis.
