Stem Cell Media

Stem cell media are specially formulated liquid culture systems that provide the nutrients, salts, growth factors, and signaling molecules needed to maintain, expand, or direct the differentiation of stem cells in vitro. Their composition regulates cell behavior by supplying metabolic substrates and activating extracellular signals that influence self-renewal, lineage commitment, survival, and proliferation, while serum or defined supplements can alter cellular responses. Researchers tailor media to the stem cell type and experimental goal, including maintenance of pluripotency, generation of specialized cell types, and organoid formation. Reliable media formulations improve reproducibility in developmental biology, disease modeling, drug screening, and regenerative research.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
