Stereotaxic Implantation

Stereotaxic implantation is a surgical technique that places electrodes, cannulas, probes, or other devices at precisely defined locations within the brain or other anatomical structures. Using a stereotaxic frame or robotic system, researchers stabilize the subject, identify anatomical landmarks, and guide an instrument along calculated three-dimensional coordinates to reach the target while limiting tissue disruption. In biology, this method supports controlled drug delivery, neural recording and stimulation, lesion studies, and behavioral experiments. Accurate implantation improves the reliability and reproducibility of measurements, helping researchers connect localized neural activity or manipulation with physiology, behavior, and disease mechanisms.

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.

This work presents a protocol to perform a stereotaxic, neurosurgical implantation of microelectrode arrays in the common marmoset. This method specifically enables electrophysiological recordings in freely behaving animals but can be easily adapted to any other similar neurosurgical intervention in this species (e.g., cannula for drug administration or electrodes for brain stimulation).
