Structural Determination

Structural determination is the process of identifying the three-dimensional arrangement of atoms in a molecule, especially proteins, nucleic acids, and biomolecular complexes. In biology, researchers infer structure by measuring how molecules interact with radiation or magnetic fields, using methods such as X-ray crystallography, nuclear magnetic resonance spectroscopy, and cryo-electron microscopy, followed by computational model building and validation. These structures reveal active sites, binding interfaces, conformational changes, and molecular interactions that explain biological function. Structural determination supports drug discovery, protein engineering, diagnosis of molecular defects, and the study of mechanisms underlying cellular processes.

β-barrel outer membrane proteins (OMPs) serve many functions within the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Here, we hope to alleviate a known bottleneck in structural studies by presenting protocols for the production of β-barrel OMPs in sufficient quantities for structure determination by X-ray crystallography or NMR spectroscopy.
