Subcutaneous Tumor

A subcutaneous tumor is an abnormal mass that develops in the tissue beneath the skin, where it may be benign or malignant and can alter local anatomy and function. Tumor formation occurs when cells acquire changes that disrupt normal controls on proliferation, survival, and tissue organization; continued growth depends on interactions with surrounding connective tissue, blood vessels, and immune cells. In biology, subcutaneous tumors provide accessible systems for examining tumor development, progression, and host responses, while clinical evaluation commonly uses physical examination and imaging, with tissue analysis helping establish cellular characteristics and guide diagnosis or research.

Evaluation of Tumor-infiltrating Leukocyte Subsets in a Subcutaneous Tumor Model

This protocol describes a method for the detailed evaluation of leukocyte subsets within the tumor microenvironment in a mouse tumor model. Chemerin-expressing B16 melanoma cells were implanted subcutaneously into syngeneic mice. Cells from the tumor microenvironment were then stained and analyzed by flow cytometry, allowing for detailed leukocyte subset analyses.

Here we describe a method to separate and enrich components of the tumor immune and non-immune microenvironment in established subcutaneous tumors. This technique allows for the separate analysis of tumor immune infiltrate and non-immune tumor fractions which can permit comprehensive characterization of the tumor immune microenvironment.

Human tumor xenografts in immunodeficient mice are valuable tools to study cancer biology. Specific protocols to generate subcutaneous and intrahepatic xenografts from human hepatocellular carcinoma cells or tumor fragments are described. Liver regeneration induced by partial hepatectomy in recipient mice is presented as a strategy to facilitate intrahepatic engraftment.
