Subpopulation Cell Cycle

Subpopulation cell cycle analysis examines how distinct groups of cells progress through the cell cycle, revealing differences in proliferation, quiescence, or arrest within a mixed population. Cells are classified by phase, typically G1, S, or G2/M, using DNA content and, when needed, markers of DNA synthesis or mitosis; comparing these distributions shows how each subpopulation responds over time or to experimental conditions. This approach supports studies of development, cancer biology, stem cells, and drug response by identifying phase-specific effects that bulk measurements can obscure, helping researchers link cellular heterogeneity to growth, differentiation, and treatment outcomes.

Analysis of Cell Cycle Position in Mammalian Cells

Determining the cell cycle position of a population of cells, or understanding how signals affect proliferation, can be readily measured by flow cytometry using this protocol. We report a simple experimental approach to staining cells and quantifying their position in the cell cycle.

This video demonstrates an in-situ fluorescence multispectral imaging technique to analyze different subpopulations of infiltrating T cells in tumor tissue. A renal cell carcinoma tissue cryosection is treated with antibodies to label CD8+ T cells that express specific inhibitory receptors and exhibit an immunotherapy-resistant phenotype. The labeled section is visualized using a multispectral microscope to identify the distinct CD8+ T cell subpopulations.
