Synergistic Activation

Synergistic activation is a biological process in which two or more signals combine to produce a response greater than the effect expected from each signal alone. It occurs when interacting receptors, signaling pathways, or regulatory factors reinforce one another, increasing signal strength, lowering response thresholds, or coordinating downstream cellular activity. This principle helps explain how cells integrate complex environmental and internal cues to control processes such as immune responses, gene expression, development, and metabolism. Studying synergistic activation can reveal how biological systems achieve sensitive, context-dependent regulation and can inform research into signaling networks, therapeutic combinations, and interventions designed to enhance or limit cellular responses.

Divide the transwell system into five groups that undergo various treatments. The first group serves as the control and consists of a co-culture of neuroblastoma and microglia cells. In the second group, treat the co-culturer with fibrillar β-amyloid (fAβ) alone. fAβ induces damage in neuroblastoma cells. In the third group, treat neuroblastoma cells with an 808-nanometer laser, then co-culture them with untreated microglia. The laser reduces oxidative stress, improving neuronal survival. In the...

Synergistic Effect of Intracranial and Intraperitoneal Delivery of Analgesic Drugs on Thermal Pain Response in a Rat

Source: Martinez, E., et al. Intracranial Pharmacotherapy and Pain Assays in Rodents. J. Vis. Exp. (2019).This video demonstrates the intracranial administration of an analgesic drug in an anesthetized rat with a pre-implanted guide cannula in the prefrontal cortex. An additional analgesic drug is co-administered intraperitoneally. Following recovery, the rat’s thermal pain response is assessed using Hargreave’s apparatus, where paw withdrawal latency is measured to evaluate the analgesic...

Eukaryotic Transcription Activators

Transcription activators are proteins that promote the transcription of genes from DNA to RNA. In most cases, these proteins contain two separate domains ‒ a domain that binds to DNA and a domain for activating transcription; however, in some cases, a single domain is responsible for both binding and activation of transcription, as seen in the glucocorticoid receptor and MyoD. The binding domains are capable of recognizing and interacting with regulatory sequences on the DNA. These domains are...

Synergistic Antimicrobial Effect of Membrane-Targeting Compounds on Antibiotic-Resistant Bacteria

Take suspensions of an antibiotic-resistant strain of a pathogenic bacterium.Treat one suspension with a short, membrane-disrupting antibiotic compound and the other with both the short compound and a longer, membrane-intercalating compound.Incubate both suspensions with agitation. These compounds mimic the lipid bilayer structure, enabling their spontaneous insertion into bacterial membranes. The short compound spans only part of the membrane’s thickness, creating a structural mismatch. This...

We show that a developed biomedical device involving continuous or pulsed visible laser based treatment that is combined with antibiotic treatment (gentamycin), results in a statistically significant synergistic effect leading to a reduction in the viability of P. aeruginosa PAO1, by 8 log's compared to antibiotic treatment alone.
