Peptide Exit Tunnel Function

Peptide exit tunnel function refers to how the ribosomal tunnel guides and influences a newly synthesized polypeptide as it emerges from the ribosome. During translation, the nascent peptide moves through this narrow passage, where tunnel geometry and interactions with ribosomal RNA and proteins can affect folding, modification, and the progression of peptide bond formation. Specific amino acid sequences may interact with the tunnel to slow or pause translation, allowing regulatory signals to control protein synthesis. Understanding this function helps explain co-translational folding, antibiotic action, and how cells regulate the production of functional proteins.

Specific short peptides derived from Nef and protein transfection were exploited to determine the structure, function, and binding partners of Nef’s Secretion Modification Region.
