Metabolite Binding

Metabolite binding is the reversible association of small molecules produced or consumed by metabolism with proteins, nucleic acids, or membranes, influencing how cells sense and use chemical resources. It occurs when complementary shape, charge, and polarity allow a metabolite to occupy a binding pocket or interact with a molecular surface; these interactions can alter protein conformation, enzyme activity, transport, or signaling. Measuring metabolite binding helps explain allosteric regulation, feedback control, metabolic flux, and competition between endogenous metabolites and drugs. In biology, binding assays, structural methods, and metabolomics connect molecular interactions to cellular phenotypes, supporting research into disease mechanisms, biomarker development, and therapeutic design.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
