Tc Tag Specificity

Tc tag specificity refers to the selective recognition of a short tetracysteine peptide sequence engineered into a protein for site-specific labeling. Biarsenical dyes such as FlAsH and ReAsH bind the cysteine-rich CCXXCC motif, becoming fluorescent when complexed with the tag, while unbound dye can be removed to reduce background signal. This approach enables researchers to track protein localization, trafficking, and dynamics in living cells with relatively small tags that may perturb protein function less than larger fluorescent proteins. Tc tag specificity is therefore valuable in cell biology, molecular imaging, and studies of protein behavior in native biological environments.

Addition of a tag to a protein is a powerful way of gaining insight into its function. Here, we describe a protocol to endogenously tag hundreds of Trypanosoma brucei proteins in parallel such that genome scale tagging is achievable.
