Temperature Sensitive Allele

A temperature-sensitive allele is a genetic variant whose effect on an organism changes with environmental temperature, allowing gene function to be conditionally controlled. At a permissive temperature, the altered protein may fold or function sufficiently to support a normal phenotype, whereas a restrictive temperature can destabilize the protein or impair its activity, revealing the mutation’s effects. These alleles help researchers investigate essential genes that cannot be studied through permanent loss of function, especially in developmental biology and cell biology. By shifting temperature at defined stages, scientists can examine when and where a gene contributes to processes such as cell division, differentiation, and tissue development.

The protocol presents the optimized parameters for preparing thermosensitive liposomes using the staggered herringbone micromixer microfluidics device. This also allows co-encapsulation of doxorubicin and indocyanine green into the liposomes and the photothermal-triggered release of doxorubicin for controlled/triggered drug release.

Genetic associations often remain unexplained at a functional level. This method aims to assess the effect of phenotype-associated genetic markers on gene expression by analyzing cells heterozygous for transcribed SNPs. The technology allows accurate measurement by MALDI-TOF mass spectrometry to quantify allele-specific primer extension products.
