Temperature Sensitivity Assay

A temperature sensitivity assay is an experimental method used to determine how changes in temperature affect the growth, survival, or biological activity of cells, organisms, or biochemical systems. In a controlled design, matched samples are exposed to defined temperatures and compared by measuring outcomes such as colony formation, viability, growth rate, or enzyme activity. The resulting temperature-response profile can reveal optimal conditions, stress thresholds, and defects in processes that depend on protein stability, membrane function, or metabolic regulation. In biology, these assays support studies of microbial physiology, genetic phenotypes, cellular stress responses, and the functional effects of temperature-sensitive mutations.

The protocol presents the optimized parameters for preparing thermosensitive liposomes using the staggered herringbone micromixer microfluidics device. This also allows co-encapsulation of doxorubicin and indocyanine green into the liposomes and the photothermal-triggered release of doxorubicin for controlled/triggered drug release.
