Tetracycline Operator Array

The Tetracycline Operator Array is a repeated DNA sequence containing tetracycline operator sites that enables controlled regulation of gene expression, making it a versatile tool in molecular and cell biology. Tet repressor proteins bind these operator sites and can block transcription; adding tetracycline or doxycycline changes repressor binding, allowing researchers to switch expression on or off, depending on the engineered Tet system. Arrays are incorporated into promoters to amplify regulatory control and support tunable, inducible expression in cultured cells, transgenic organisms, and functional studies. This conditional control helps investigate gene function, cellular pathways, development, and disease-related mechanisms.

A rapid and simple way to generate human cell lines with inducible and reversible cDNA overexpression or shRNA-mediated knock-down of the gene of interest. This method enables researchers to reliably and highly reproducibly manipulate cell lines that are difficult to alter by transient transfection methods or conventional knockdown/knockout strategies.
