Theca Folliculi

Theca folliculi are specialized layers of ovarian stromal cells that surround developing follicles and support follicle growth, hormone production, and ovulation. The inner theca interna responds primarily to luteinizing hormone by producing androgen precursors, which granulosa cells convert into estrogens under follicle-stimulating hormone stimulation, while the outer theca externa provides structural and vascular support. Studying theca folliculi helps explain folliculogenesis, ovarian steroidogenesis, and the hormonal coordination of reproductive cycles. Changes in theca-cell function are also relevant to conditions such as polycystic ovary syndrome and to research on fertility, ovarian physiology, and assisted reproduction.

Here, we describe a protocol for separating yolk, granulosa cells, and theca cells in avian preovulatory follicles. This precision handling enables critical investigations into the role of these layers in reproductive function, aiding the understanding of follicular development, hormonal regulation, and disease research for enhanced agricultural yield and biomedical insights.
