Tissue Degeneration

Tissue degeneration is the progressive loss of cellular structure, function, or viability within a tissue, often impairing organ performance and contributing to disease. It can arise when cellular damage, inflammation, oxidative stress, impaired repair, or disrupted signaling exceeds the tissue’s ability to maintain homeostasis and replace injured cells. In biology, studying tissue degeneration helps researchers connect molecular and cellular changes with altered tissue architecture and physiological decline. Experimental models, histological analysis, imaging, and molecular assays are used to characterize these processes, evaluate potential treatments, and investigate conditions affecting the nervous system, muscles, joints, and internal organs.

Sea lamprey lose the gall bladder and bile ducts during metamorphosis, a process similar to human biliary atresia. A new fixation and clarification method (CLARITY) was modified to visualize the entire biliary tree using laser scanning confocal microscopy. This method provides a powerful tool to study biliary degeneration.

Intervertebral disc degeneration is a significant contributor to back pain and a leading cause of disability worldwide. Numerous animal models of intervertebral disc degeneration exist. We demonstrate an ovine model of intervertebral disc degeneration, utilizing a drill bit, which achieves a consistent disc injury and reproducible level of disc degeneration.
