Tissue Oxygen

Tissue oxygen is the oxygen available within body tissues to support cellular metabolism, energy production, and normal physiological function. Oxygen enters the circulation through the lungs, binds reversibly to hemoglobin in red blood cells, and reaches tissues through blood flow; it then diffuses down a partial-pressure gradient from capillaries into cells, where mitochondria use it for aerobic respiration. Measuring or modeling tissue oxygen helps biologists investigate how oxygen delivery changes with metabolism, blood flow, and tissue structure. These principles are central to understanding hypoxia, wound healing, tumor biology, exercise responses, and the effects of vascular or respiratory disorders.

An Assay for Detecting Reactive Oxygen Species Production in Rice Tissues upon Immune Elicitation

This video demonstrates an assay for real-time detection of reactive oxygen species (ROS) production in rice tissues upon pathogen-associated molecular pattern (PAMP) elicitation. Cut sheath segments and leaf discs are exposed to a bacterial flagella-derived peptide functioning as a PAMP. Upon recognition of the peptide via pattern recognition receptors (PRRs) on plant cells, a rapid and transient production of ROS occurs, which is detected via a chemiluminescence assay.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

Measurement of Tissue Oxygenation Using Near-Infrared Spectroscopy in Patients Undergoing Hemodialysis

We present a protocol to measure regional oxygen saturation (rSO2) in hemodialysis (HD) patients by using a near-infrared spectroscopy monitor. The rSO2 value is an index of tissue oxygenation. This noninvasive and real-time monitoring could be useful for confirming changes in organ oxygenation during HD.
