Membrane Attack Complex

The membrane attack complex (MAC) is a terminal structure of the complement system that protects against pathogens by damaging target cell membranes. Complement proteins C5b through C9 assemble sequentially on a membrane, forming a ring-like pore that disrupts membrane integrity, causes ion imbalance, and can lead to cell lysis. MAC activity is especially important in defense against susceptible bacteria, while host cells use regulatory proteins to limit unintended complement damage. Studying MAC assembly and regulation helps explain immune deficiencies, inflammatory disease, and pathogen evasion, and supports research into complement-targeted therapies.
