Tmp Uv Mutagenesis

TMP-UV mutagenesis is a laboratory method for creating genetic variation by treating cells with trimethylpsoralen (TMP) and ultraviolet A light, making it useful for studying gene function and DNA repair. TMP intercalates between DNA bases, and UV-A activation causes covalent links between pyrimidines on opposing DNA strands. These crosslinks disrupt replication and transcription, while repair and lesion-bypass processes can introduce mutations when cells recover. Researchers use the method in forward genetic screens to identify genes involved in biological pathways, stress responses, and genome maintenance. Resulting mutants can reveal gene functions and provide models for investigating mutation, recombination, and DNA damage responses.
