Transcriptional Profiling

Transcriptional profiling is the systematic measurement of RNA transcripts in a cell, tissue, or organism to characterize gene activity under specific conditions. The process typically involves isolating RNA, converting it to complementary DNA, and quantifying transcript abundance through methods such as RNA sequencing or microarray hybridization; comparing profiles can reveal differentially expressed genes. In biology, transcriptional profiling helps define cell states, trace responses to environmental or developmental signals, identify disease-associated pathways, and evaluate experimental treatments. By linking changes in gene expression to cellular functions, it provides a foundation for studying regulation, phenotype, and biological mechanisms.

Here we present a protocol for laser-assisted microdissection of specific plant cell types for transcriptional profiling. While the protocol is suitable for different species and cell types, the focus is on highly inaccessible cells of the female germline important for sexual and apomictic reproduction in the crucifer genus Boechera.
