Vascular Smooth Muscle

Vascular smooth muscle is the contractile tissue in the walls of arteries and veins that regulates blood vessel diameter, blood flow, and vascular resistance. When signaling molecules or changes in pressure raise intracellular calcium, calcium binds calmodulin and activates myosin light-chain kinase, enabling actin-myosin interactions that constrict or relax the vessel. In biology and biomedical research, studying vascular smooth muscle helps explain blood pressure control, tissue perfusion, and disorders such as hypertension and atherosclerosis. Its responses to hormones, neurotransmitters, and mechanical forces also support research into cardiovascular therapies and vascular tissue engineering.

The purpose of this protocol is to demonstrate the isolation and culture techniques of murine primary vascular smooth muscle cells (VSMCs) from the coronary circulation. Once VSMCs have been isolated, they can be used for many standard culture techniques.

Calcification of Vascular Smooth Muscle Cells and Imaging of Aortic Calcification and Inflammation

2016

Vascular calcification is an important predictor of and contributor to human cardiovascular disease. This protocol describes methods for inducing calcification of cultured primary vascular smooth muscle cells and for quantifying calcification and macrophage burden in animal aortas using near-infrared fluorescence imaging.

Measuring Proliferation of Vascular Smooth Muscle Cells Using Click Chemistry

2019

Proliferation is a critical part of cellular function, and a common readout used to assess potential toxicity of new drugs. Measuring proliferation is, therefore, a frequently used assay in cell biology. Here we present a simple, versatile method of measuring proliferation that can be used in adherent and non-adherent cells.

Extraction and Purification of Polyphenols from Freeze-dried Berry Powder for the Treatment of Vascular Smooth Muscle Cells In Vitro

This work details a step-by-step method to prepare polyphenol-rich extracts from freeze-dried berry powder. In addition, it provides a thorough description of how to use these polyphenol-rich extracts in cell culture in the presence of the peptide hormone angiotensin II (Ang II) using Vascular Smooth Muscle Cells (VSMCs).

Von Kossa Staining of Aortic Vascular Smooth Muscle Cells: An In Vitro Technique for Assessment of Arterial Medial Layer Calcification
