Vector Shield Mounting

Vector Shield mounting is a fluorescence microscopy method that preserves labeled biological specimens beneath a coverslip using an antifade mounting medium. The medium surrounds the sample, limits photobleaching by reducing light-induced chemical damage to fluorophores, and helps maintain signal during image acquisition. In biology, this preparation supports immunofluorescence, histology, and cellular imaging by improving the stability and visibility of fluorescent labels in fixed tissues or cultured cells. Consistent mounting also helps produce clearer images and more reliable comparisons across specimens, making it useful for examining protein localization, tissue organization, and cell morphology with widefield or confocal microscopy.

The protocol described here enables researchers to specifically modify adenovirus capsids at selected sites by simple chemistry. Shielded adenovirus vectors particles and retargeted gene transfer vectors can be generated, and vector host interactions can be studied.
