Viral Gene Amplification

Viral gene amplification is the production of many copies of a viral DNA or RNA sequence, either during infection or through laboratory methods, enabling genetic material to be detected and studied. In the laboratory, reverse transcription can convert viral RNA into complementary DNA, after which repeated cycles of primer binding and DNA synthesis, as in polymerase chain reaction, exponentially increase a target region; amplification of DNA viruses can begin directly from DNA. This approach supports sensitive detection of viral infection, measurement of viral genetic material, sequencing, mutation analysis, and characterization of viral diversity, making it valuable in diagnostics, epidemiology, and basic virology research.

We describe the usage of a fluorescent reporter vaccinia virus that enables real-time measurement of viral infectivity and gene expression through the stage-specific expression of spectrally distinct reporter fluorophores. We detail a plate-based method for accurately identifying the stage at which virus replication is affected in response to small molecule inhibition.
