Viral Rna Isolation

Viral RNA isolation is a laboratory technique used to extract intact viral ribonucleic acid from biological samples for downstream analysis in biology and biomedical research. The process typically disrupts viral particles, inactivates contaminating ribonucleases with chaotropic reagents, and captures RNA on silica membranes or magnetic beads before washing and elution; careful handling and RNase-free conditions help preserve RNA quality. Purified viral RNA supports reverse transcription polymerase chain reaction, sequencing, viral detection, genome characterization, and studies of infection and transmission. Reliable isolation improves assay sensitivity and enables researchers to compare viral variants, monitor outbreaks, and investigate host-pathogen interactions.

Detection of Viral RNA by Fluorescence in situ Hybridization (FISH)

A fluorescence in situ hybridization (FISH) method was developed to visually detect viral genomic RNA using fluorescence microscopy. A probe is made with specificity to the viral RNA that can then be identified using a combination of hybridization and immunofluorescence techniques. This technique offers the advantage of identifying the localization of the viral RNA or DNA at steady-state, providing information on the control of intracellular virus trafficking events.

This method describes the use of click chemistry to measure changes in host cell transcription after infection with the Rift Valley fever virus (RVFV) strain MP-12. Results can be visualized qualitatively via fluorescence microscopy or obtained quantitatively through flow cytometry. This method is adaptable for use with other viruses.

Isolation and Characterization of RNA-Containing Exosomes

This paper demonstrates methods for the isolation, purification and detection of exosomes, as well as techniques for analysis of their molecular content. These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.
