Zebrafish Microinjection

Zebrafish microinjection is a laboratory technique that delivers precise amounts of genetic, chemical, or cellular material into zebrafish embryos, making it valuable for studying development and gene function. Under a microscope, a fine glass micropipette penetrates the chorion and deposits material into the yolk, cytoplasm, or a developing cell, often at the one-cell stage; injected contents can include mRNA, DNA, morpholinos, CRISPR reagents, or fluorescent tracers. Researchers use this method to manipulate gene expression, generate transgenic or mutant models, trace cell lineages, and investigate developmental processes, disease mechanisms, and potential therapeutic strategies.

One of the major advantages to working with zebrafish (Danio rerio) is that their genetics can be easily manipulated by microinjection of early stage embryos. Using this technique, solutions containing genetic material or knockdown constructs are delivered into the blastomeres: the embryonic cells sitting atop the yolk of the newly fertilized egg. Delivery into the cytoplasm is achieved either through direct injection into the blastomere, or via natural cytoplasmic movements that occur after a...

Renal injuries incurred from nephrotoxins, which include drugs ranging from antibiotics to chemotherapeutics, can result in complex disorders whose pathogenesis remains incompletely understood. This protocol demonstrates how zebrafish can be used for disease modeling of these conditions, which can be applied to the identification of renoprotective measures.
